Melanotan II (MT-2) is a synthetic cyclic analog of alpha-melanocyte-stimulating hormone (α-MSH). Originally developed by researchers at the University of Arizona as a potential preventive treatment for skin cancer, theorizing that increased melanin could protect against UV damage.

MT-2 selectively binds to the melanocortin receptor MC1R, which regulates the conversion of tyrosine into eumelanin — a brown-to-black pigment that effectively absorbs UV rays and disperses them in the skin’s surface layers, actively protecting epidermal cells from oxidative stress.

MT-2 also activates MC4R receptors expressed in the hypothalamus and other CNS regions, modulating sexual arousal, appetite, and energy expenditure. Research published in the International Journal of Molecular Sciences indicates this MC4R activation influences the neuroendocrine regulation of hunger and satiety.
